(1) Drug properties: The compressibility and crystal shape of the drug have a direct impact on the difficulty of tablet molding. Compressibility refers to the plasticity of drug particles during compression.
Generally, the material with large plastic deformation can form a stable tablet after being pressed, and it can be formed without adding auxiliary materials and adhesives. The elastic deformation of the drug is large. When the pressure is released, the elastic recovery phenomenon sometimes occurs, resulting in splits or loose pieces.
To overcome elastic recovery, adhesive must be added. The viscosity of the adhesive depends on the strength of the elastic deformation of the drug.
(2) Crystallization water and water content: an appropriate amount of crystal water or water in granular powder is the most important factor for tablet molding, which increases the plasticity of drug particles and reduces elasticity; The surface forms a thin layer. It is convenient for the particles to approach each other and generate sufficient cohesion.
(3) Binders and lubricants: Medicines with poor compressibility are difficult to form, and a binder must be added to increase the cohesion of the particles to facilitate the forming of the tablet.
Generally, hydrophilic polymer compounds with higher cohesion are added. After mixing with the drug, it can be granulated by dry or wet granulation. Lubricants reduce friction and increase granule flow, resulting in compact tablet compression.
